Description

Siglec-11 is a 686 amino acid single-pass type I membrane protein that contains one Ig-like V-type domain and three Ig-like C2-type domains. In humans, Siglec-11 is widely expressed on tissue macrophages, which include microglia in the brain. No expression of Siglec-11 is seen in the brain compartment of non-human primates. Siglec-11 functions as an adhesion molecule that preferentially binds to α-2, 8-linked sialic acid trimers and mediates sialic acid binding to cells. It is a negative regulator of microglia, both related to inflammation and phagocytic function. In murine microglial cells with ectopic expression of human Siglec-11, stimulation of Siglec-11 could induce the transcription of the pro-inflammatory mediator genes, such as IL-1β and nitric oxide synthase-2. Phagocytosis of apoptotic neuronal material has shown to be reduced in Siglec-11 expressing microglia. Co-culture of Siglec-11 expressing microglia and neurons demonstrated neuroprotective functions of Siglec-11.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Function
Adhesion molecule that mediates sialic-acid dependent binding to cells. The cytosolic domain of Siglec-11 is phosphorylated at tyrosine residue(s) upon pervanadate treatment of cells and then it recruits the protein-tyroisine phosphatases SHP-1 and SHP-2. Such interaction induces inhibitory cellular signals in immune response.
