Total cell lysate from human HepG2 cells (lane 1) and mouse liver cells (lane 2) were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified monoclonal anti-NR5A2 (clone 1D12B67) antibody. Proteins were visualized using an HRP-conjugated anti-mouse-IgG secondary and chemiluminescence detection.

Description

The nuclear receptor subfamily 5 group A member 2, also known as NR5A2, is a 54 kD nuclear receptor protein. NR5A2 is a member of the FTZ-F1 sub-family and is highly expressed in the pancreas and to a lesser extent in the liver. NR5A2 contains a DNA binding domain, a hormone ligand-binding domain, and two zinc finger domains. It also binds to DNA as a monomer. NR5A2 is known to regulate the transcription of the liver enzyme cholesterol 7-alpha-hydrolase, which converts cholesterol to bile acids and also activates the hepatitis B virus enhancer II to contribute to the liver-specific expression of this virus. NR5A2 has been suggested to regulate pancreas-specific gene expression and to play a role in embryonic development. There are at least 3 alternatively spliced isoforms of NR5A2: isoforms 1 and 2 are present in fetal and adult liver and in liver cancer cells HepG2 as well.

Antigen Details

Structure
Orphan nuclear receptor, member of the FTZ-F1 sub-family, contains a DNA binding domain, 2 zinc finger motifs, a FTZ-F1 box, and a ligand-binding domain. Approximately 54 kD, three alternatively spliced variants exist.
Distribution
Nuclear, highly expressed in the pancreas, lower expression in the liver, and very low expression in the heart and lungs.
Function
Binds DNA as a monomer to regulate the transcription of the liver enzyme cholesterol 7-alpha-hydroxylase, which converts cholesterol to bile acids. Activates viral hepatitis B enhancer II to regulate liver-specific expression of this virus in combination with other transcription factors.
Interaction
Interacts wtih GRIP1, NCOA2, and NR0B2.
