Brief Protocol: |
Resuspend in PBS/BSA and count the cells. Load up to 5x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (24.62) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl of antibody staining solution containing CD11b Pacific Blue™(1:1600). Incubate 15min at 4°C in the dark. Wash with PBS/BSA, spin down resuspend and transfer to FACS tubes to read. Use PI to exclude the dead cells. |
Results Summary: |
Macrophages are higher expressors of CD11b, while B cells (B-1 cells) that are enriched in the peritoneal cavity are intermediate expressors of CD11b. |
Additional Notes: |
This antibody is very good to sort macrophages and B cells from the peritoneal cavity. |
