Brief Protocol: | Diluted GAM 4053 at 1:1000, 1:5000, and 1:15000 and coated on 96 well plate with bicarbonate buffer. 75 µL used to coat wells (1 hr) Performed serial dilutions 1:5 of target epitope (mouse IgG1) starting at 2250 ng, down to 3.6 ng per well. Used cRPMI as negative control. (1 hr) Washed, then Used 2 different dilutions of detection antibody (Invitrogen cat# 62-6520), for each dilution of capture antibody. (1 hr) Washed, then added quanta blue fluorescing substrate (30 min), quenched, and read plate on fluoresence plate reader. |
Results Summary: | 1:1000 dilution gave best results with 1:8000 dilution of detection mab. Produced a nice log curve. |
