Total lysates (15 µg protein) from Raw 264.7 (Lane 1) and Jurkat (Lane 2) cells were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 1 µg/mL purified anti-NFATc2 antibody, clone 16A12A31 (upper). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-mouse-IgG secondary antibody conjugated to HRP for the anti-NFATc2 Antibody or 1:5000 diluted Direct-Blot™ HRP anti-β-Actin antibody, clone 2F1-1 (lower). Description

NFATc2 is a family member of the NFAT (calcineurin-dependent nuclear factor of activated T cells) transcription factors, regulating expression of the genes involved in T-cell activation and proliferation. In resting T cells, NFATc2 protein is phosphorylated and is present in the cytoplasm. Upon stimulation, NFATc2 protein is dephosphorylated by calcium dependent phosphatase, resulting in the translocation of NFATc2 protein to the nucleus, where it induces expression of target genes. In addition to its roles in immune cells, NFATc2 is revealed to mediate VEGF signaling, transactivating the gene encoding cyclooxygenase 2 (COX2). The production of prostaglandins by cycloxoygenase 2 is required for the migration of endothelial cells and angiogenesis. NFATc2 is also suggested to participate in tumorigenesis and invasive migration of tumor cells.
