Whole cell extracts (15 µg protein) from HeLa cells untreated (-) or treated (+) with 300 ng/mL nocodazole overnight were resolved on a 4-12% Bis-Tris gel, transferred to nitrocellulose and probed with 0.0625 µg/mL (1:8000 dilution) of purified anti-MEK1 Phospho (Ser298) Antibody, clone A16117B, for 2 hours at room temperature. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Equal MEK1 loading was confirmed by probing membranes with an anti-pan MEK1 antibody. Lane M: Molecular Weight marker.

Description

MEK1 and MEK2 (Map/Erk Kinase) are serine/threonine kinases in the MAPK/ERK signaling axis that play a critical role in cellular proliferation, differentiation, and migration. Phosphorylation at MEK1 (Ser298) by PAK1 enhances the ability of upstream Raf kinases to phosphorylate regulatory serines (Ser218 and Ser222) in the activation loop, thereby stimulating kinase activity.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.0625 - 1.0 µg per mL. For flow cytometry using our True-Phos™ Perm Buffer, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
