A single cell suspension from C57BL/6 mouse bone marrow was prepared for the positive selection of CD117 (c-Kit)+ cells using the MojoSort™ Mouse CD117 (c-Kit) Selection Kit. The cells were incubated with TruStain FcX™ (anti-mouse CD16/32) antibody prior to the addition of the anti-mouse CD117 (c-Kit) PE antibody. After separation, the cells were stained with APC anti-mouse CD117 (c-Kit) (clone ACK2) and Brilliant Violet 421™ anti-mouse lineage cocktail (clones 17A2/RB6-8C5/RA3-6B2/Ter-119/M1/70). Dead cells were excluded by 7-AAD. -
A single cell suspension from C57BL/6 mouse bone marrow was prepared for the positive selection of CD117 (c-Kit)+ cells. The MojoSort™ Mouse Hematopoietic Progenitor Cell Isolation Kit (Cat. No. 480003/480004) was first used to remove Lin+ cells. The MojoSort™ Mouse CD117 (c-Kit) Selection Kit was then used to further enrich for CD117 (c-kit)+ cells. The cells were incubated with TruStain FcX™ (anti-mouse CD16/32) antibody prior to the addition of the PE anti-mouse CD117 (c-Kit) antibody. After separation, the cells were stained with APC anti-mouse CD117 (c-Kit) (clone ACK2) and Brilliant Violet 421™ anti-mouse lineage cocktail (clones 17A2/RB6-8C5/RA3-6B2/Ter-119/M1/70). Dead cells were excluded by 7-AAD.

Mouse CD117 (c-Kit)+ cells are either selected or depleted by incubating the sample with a PE conjugated anti-mouse CD117 (c-Kit) antibody and then magnetic Mouse anti-PE Nanobeads. The magnetically labeled fraction is retained by the use of a magnetic separator. After collection of the CD117 (c-Kit)+ expressing cells, downstream applications include functional assays, gene expression, phenotypic characterization, etc.
MojoSort™ reagents are also compatible with column-based cell separation systems available from other vendors. Optimized protocols for cell separation using columns from in-house testing are provided for each kit under the “Related Protocols” section, as well as representative data on the product webpage (where available). Data generated using column separators are indicated on the figure legend.
Due to the property of the beads, MojoSort™ reagents typically require dilution for optimal use on column separators. Where available, recommended dilution factors for each kit component based on in-house testing are provided under the “Application Notes” section of the webpage.

MojoSort™ magnetic particles can be used with other commercially available magnetic separators, both free standing magnets and column-based systems. Because MojoSort™ protocols are optimized for the MojoSort™ separator, the protocols may need to be adjusted for other systems. Please contact BioLegend Technical Service for more information and guidance. We do not recommend using MojoSort™ particles for BD’s IMag™ or Life Technologies’ DynaMag™.
