Naïve CD4 T cells were harvested from the spleens of Il22+/+ or Il22-/- mice (C57BL/6 background, see Immunity. 2007 Oct; 27(4):647-59).
Cells were then cultured with plate-bound aCD3 and aCD28 mAbs in IMDM (Th0 Conditions); or additionally with recombinant mouse IL-23 (50 ng/ml), IL-1b (20 ng/ml), IL-6 (20 ng/ml) and 200 nM 6-formylindolo(3,2-b)carbazole (Th22 Conditions).
Three days post-stimulation, supernatants were harvested, and IL-22 was quantitated using the LEGEND MAX™ Mouse IL-22 ELISA Kit (Cat. No. 436307).

IL-22 belongs to a family of cytokines structurally related to IL-10, including IL-19, IL-20, IL-24, and IL-26. Mouse IL-22 shares 79% amino acid identity with human IL-22. IL-22 is produced by immune cells and acts on nonimmune cells to regulate local tissue inflammation. As a product of the T helper 17 lineage of CD4+ effector lymphocytes, IL-22 plays a critical role in mucosal immunity as well as in the disregulated inflammation observed in autoimmune diseases.

We typically use a stabilizer for pre-coated plates. The additional washing step is designed to remove these components before you start the assay. If you do not perform the washing, the effect on assay performance is negligible.
- I have multiple LEGEND MAX™ ELISA kits that I want to run simultaneously. Can I use the same wash buffer for all the kits?

The wash buffer provided in all our LEGEND MAX™ kits is the same and the part numbers on the wash buffer bottles in these kits should be identical. For ELISA MAX™ Deluxe and ELISA MAX™ Standard Sets, we provide a recipe for the wash buffer on each kit’s technical data sheet. This recipe is the same for all ELISA MAX™ sets.
- For some of your ELISA kits, why do my serum samples require dilution with assay buffer?

In some cases, dilution with assay buffer is required to minimize the matrix difference between the samples and the standards to achieve better accuracy.
