IHC staining of HRP anti-Gelsolin antibody (clone CPTC-Gelsolin-2) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 5 µg/mL of the primary antibody overnight at 4°C. DAB was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

Description

After severing, gelsolin remains bound to the barbed end of the actin filament, preventing the reannealing of severed filaments. Gelsolin is crucial for the maintenance of actin homeostasis. Cells from gelsolin null mice exhibit a variety of defects, such as pronounced stress fibers formation, defective chemotaxis, and wound healing. Dysfunction of Gelsolin has been indicated in multiple diseases include cancer, infection, inflammation, cardiac injury, pulmonary disease, and neurodegeneration. Gelsolin is proteolytically cleaved and rendered inactive in Alzheimer’s disease (AD). Overexpression of gelsolin significantly reduces amyloid beta load in brains of AD transgenic mice.
