Total lysates (15 µg protein) from HeLa (human), CHO (hamster) and COS-7 (monkey) cells were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:1000 anti-Insulin Degrading Enzyme antibody, clone 9B12.225. Proteins were visualized using chemiluminescence detection by incubation with HRP Goat anti-Mouse secondary antibody (Cat. No. 405306, 1:3000 dilution). Direct-Blot™ HRP anti-β-actin antibody was used as a loading control (Cat. No. 643807, 1:8000 dilution).

    HeLa cells were fixed with 4% paraformaldehyde (PFA) for 15 minutes, permeabilized with 0.5% Triton X-100 for 3 minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with (A) mouse IgG1, κ isotype control (Negative, Cat. No. 401402) or (B-D) Insulin Degrading Enzyme primary antibody (Clone 9B12.225) overnight at 4°C followed by Alexa Fluor® 488 (Green) Goat anti-Mouse IgG (Cat. No. 405319) incubation for one hour at room temperature. Nuclei were counterstained with DAPI (Blue, Cat. No. 422801). The image was captured with a 60X objective using KEYENCE BZ-X700 fluorescence microscope. Exposure time (Seconds) for (A-D) is 1/12. Concentrations for (A-B) is 1:100 dilution, (C) is 1:250 dilution and (D) is 1:500 dilution.

Description

Insulin degrading enzyme (IDE), also known as insulinase, insulin protease or insulysin, cleaves the peptide hormone insulin with consequences for insulin response and resistance. Since its discovery, IDE has been shown to degrade a variety of bioactive peptides. Recently the finding that IDE clears intracellular and extracellular amyloid products has spurred research into the link between IDE function and human neurodegenerative disease such as Alzheimer’s.

Concentration
The concentration is not quantified as this product is sold as undiluted crude mouse ascites fluid. The concentration might vary from lot-to-lot and an estimated concentration would be 1-3 mg/ml.
Storage & Handling
Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody. For long-term storage, keep the antibody at -80°C.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is a dilution of 1:1000. For immunocytochemistry, a dilution range of 1:250-1:500 is recommended. For immunoprecipitation, the suggested use of this reagent is a dilution of 1:150. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody only recognizes isoform 1 at approx. 118 kD, and does not recognize isoform 2 at approx. 54 kD.

This antibody is effective in immunoblotting, immunofluorescence (IF), and immunoprecipitation (IP).

This antibody does not react with mouse.
