    C57BL/6 mouse frozen brain section was fixed with 4% paraformaldehyde (PFA) for 10 minutes at room temperature, permeabilized with 0.5% Triton X-100 for ten minutes and blocked with 5% FBS for 30 minutes at room temperature. Then the section was stained with 10 µg/mL of anti-GFAP (clone 2E1.E9) purified overnight at 4°C, followed by 2.5 µg/mL of anti-mouse IgG2b (clone RMG2b-1) Alexa Fluor® 647 (red) for 1 hours at room temperature. Nuclei were counterstained with DAPI (blue). The image was scanned with a 10X objective and stitched with the MetaMorph® software.

    IHC staining of Alexa Fluor® 647 anti-mouse IgG2b antibody on formalin-fixed paraffin-embedded human tonsil. Following antigen retrieval using Tris-EDTA, pH 9.0, the tissue was incubated with 2.5 µg/mL of purified mouse anti-CD45RA (Cat. No. 304102) at 4°C. This was followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG2b (clone RMG2b-1) at 10 µg/mL. Nuclei were counterstained with DAPI (blue, panel A) (Cat. No. 422801) and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scale: 50 µm

Description

The RMG2b-1 monoclonal antibody reacts with immunoglobulin G2b (IgG2b) in all tested mouse haplotypes (Igh-a and b). It does not react with other isotypes. The RMG2b-1 monoclonal antibody may be used as primary or secondary reagent for ELISA or immunofluorescent analysis.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. For immunohistochemistry on frozen tissue sections, a concentration range of 2.5 - 10 µg/mL is suggested. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 647 has a maximum emission of 668 nm when it is excited at 633 nm / 635 nm.
