Absolutely Necessary to Block Non-specific Antibody Binding Through FcRs (TruStain fcX™)

Significantly reduces non-specific FcR binding of flow antibodies.

Experimental Design

Application:	Flow cytometry
Cells used:	Peripheral blood leukocytes from female C57Bl/6 mouse
Brief Protocol:	Collected 25 uL blood. Washed once in cold PBS. Lysed red blood cells. Washed once in cold PBS. Stained cells in 100 uL cold PBS containing Live/Dead and TruStain Fc block (2 ug/mL) on ice for 15 minutes. Added 100 uL Thy1.1 APC (4 ug/mL) and incubated on ice for 15 minutes. Washed in cold PBS. Fixed in 100 uL 1.5% PFA until analysis.
Results Summary:	TruStain completely blocked non-specific binding to Thy1.1 APC, which B6 mice do not express.
Additional Notes:	All plots show live single cells based on live/dead exclusion. Left contour plot shows staining of peripheral blood leukocytes with Thy1.1 APC. Middle contour plot shows staining of peripheral blood leukocytes with TruStain plus Thy1.1 APC. Right contour plot shows overlay of peripheral blood leukocytes stained with Thy1.1 APC (red) or TruStain plus Thy1.1 APC (black).
