Brilliant Violet 510™ anti-human CD14 Works Well!

CD14-BV510 works great to detect monocytes in human PBMC samples.

Experimental Design

Application:	Fluorescence cytometry
Cells used:	Cryopreserved human PBMC
Brief Protocol:	Cyropreserved human PBMC were thawed, washed twice in PBS, and stained with various volumes of CD14-BV510 in a single color titration. 1 million cells in 200uL PBS were incubated with CD14-BV510 for 15 minutes at room temperature. 1uL CD14-BV510 yielded the highest stain index, however, 0.5uL detected the same frequency of monocytes with only a slightly lower MFI and we have chosen to use that volume going forward to conserve costs since we are only using it in our dump channel to gate out monocytes.
Results Summary:	0.5uL CD14-BV510 works great to detect human monocytes.
Additional Notes:	It is evident in the attached figure that CD14-BV510 clearly marks monocytes, which have higher side scatter properties than other PBMC subsets.
