Easy detection of CRTH2-positive cells within a CD4-population.

Experimental Design

Application:	Flow Cytometry
Cells used:	Human whole blood
Brief Protocol:	1. 80 µl EDTA blood plus 20 µl antibody cocktail (in FACS Buffer), incubate for 30 min at RT. 2. Lysis of erythrocytes by adding 1 ml of Lysis Buffer, incubate for 10 min at RT. 3. Wash two times with PBS; centrifuge at 400 x g for 5 min, trash supernatant. 4. Resuspend cells in 200-300 µl FACS Buffer, FACS analysis. 5. CD294-PE/Texas Red was titrated and applied at a final dilution of 1:50.
Results Summary:	Usually PE/Texas Red is very bright and can be used highly diluted, but from this antibody you need quite a bit.
Additional Notes:	Gating strategy: Lymphocytes > single cells > CD4+ > CD294+
