BAFF in B-Cell Signaling

B cell-activating factor (BAFF) can be produced by dendritic cells and T cells, and works together with BCR signaling to maintain mature B cell populations within lymphoid organs. BAFF binds several different receptors belonging to the tumor necrosis factor receptor family. Signaling through BAFFR (BR3) can activate both the classical and alternative NF-κB pathways, which will promote cell survival. Other BAFF receptors include transmembrane activator and calcium-modulating/cyclophilin ligand-interacting protein (TACI), which is expressed on all B cells, and B cell maturation Ag (BCMA), which is expressed on plasma cells. Both of these utilize TRAF signaling pathways to initiate transcription by NFAT, NF-κB, c-Jun and c-Fos, and Elk1.
