Chemokines are a family of proteins noted for their ability to induce chemotaxis and movement in cells. Most chemokines are named according to a conserved string of residues that contributes to their overall structure (CXC, CC, CX3C). Chemokines, and their receptors, are involved in a medley of immunological functions. While some are involved in migration, others play a role in inflammatory cell recruitment, virus survival, or homeostasis. Recently, experiments have elucidated the mechanism of T cell homeostasis and migration to the thymus.
T cells develop in the thymus. However, the thymus does not have hematopoietic cells, and T cells do not originate there. T cell progenitors migrate to the thymus after leaving the bone marrow or fetal liver. Recently, Calderón and Boehm shed light on chemokine receptors specifically required for entry into the thymus. Chemokines produced by thymus cells pull migrating T progenitor cells toward it. CCR7 recognizes CCL19 and CCL21, CCR9 binds CCL25, and CXCR4 responds to CXCL12. Each receptor shares some redundant function in T cell recruitment to the thymus. Deficiencies in any of these receptors, particularly CCR9, resulted in a drastic reduction in thymal T cell populations. The few T cells that were able to enter the thymus showed normal maturation and development. This work clarifies the important role of CCR7, CCR9, and CXCR4 in thymus seeding and the generation of a peripheral T cell population.
