Whole cell extracts (15 µg protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.1 µg/mL (1:5000 dilution) of purified anti-PP2C-α antibody, clone P87F12H6, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker.

Description

Protein Phosphatase 2Cα (PP2C-α) is a ubiquitously expressed magnesium- and manganese-dependent enzyme with a broad specificity of target substrates. It functions by dephosphorylating serine/threonine residues on its target proteins, and has been shown to regulate SMAD, PP2C-α, MAPK, p38 and JNK kinase signaling pathways. PP2C-α deficiency is associated with pancreatic and prostate cancers.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

When tested for western blot application, this clone produced a non-specific band of roughly ~48 kD in certain cell lines.
