Western blot of purified anti-Catalase antibody (Poly28607). Lane 1: Molecular weight marker; Lane 2: 5 µg of human liver lysate; Lane 3: 20 µg of mouse liver lysate. The blot was incubated with 0.5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled donkey anti-rabbit IgG (Cat. No. 406401). Enhanced chemiluminescence was used as the detection system.

IHC staining of purified anti-Catalase antibody (Poly28607) on formalin-fixed paraffin-embedded human liver tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 1 µg/mL of the primary antibody overnight at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

Description

Catalase is a heme-containing homotetrameric protein that decomposes hydrogen peroxide to water and oxygen to prevent the generation of hydroxyl radical in cells. The role of catalase in cellular oxidative stress defense has been widely studied. Overexpression of catalase makes cells more resistant to  hydrogen peroxide-induced toxicity and oxidant-mediated injury from hypoxia. Transgenic mice overexpressing catalase are protected against myocardial injury following adriamycin treatment. Although knockout mice of catalase develop normally, they show differential sensitivity to oxidant tissue injury. Brain is particularly vulnerable to oxidative stress. Abnormalities and dysfunction of catalase have been indicated in Parkinson’s disease, Alzheimer’s disease and others. Data show that in parkinsonian brain, catalase activity is reduced in the substantia nigra and putamen. In vitro cellular models show aggregated Abeta has high affinity for catalase, which leads to the accumulation of hydrogen peroxide and increased oxidative stress.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 1.0 µg per mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
