    Western blot of purified anti-alpha β crystalline antibody (clone Poly9079). Total tissue lysates (15 µg total protein) from human brain (lane 1), mouse brain (lane 2), and rat brain (lane 3) were resolved by 4-12% Bis-Tris gel electrophoresis, and then transferred to a PVDF membrane. The membrane was probed with 1.0 µg/mL of primary antibody overnight at 4°C, followed by incubation with HRP donkey anti-rabbit IgG antibody (Cat. No. 406401) at a 1:3000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular weight marker.

Description

Alpha B Crystallin (CryAB) is a member of the small heat shock protein (HSP20) family.  Alpha crystallins are composed of two gene products: alpha-A and alpha-B, for acidic and basic, respectively. Alpha-A is preferentially restricted to the lens and alpha-B is expressed widely in many tissues and organs. CryAB can be induced by heat shock, ischemia and oxidation. The protein is known to protect cells from stress damage. It may also play an important role in the Golgi reorganization during cell division. CryAB is also known to be associated with a number of neurodegenerative diseases such as Alzheimer’s disease and has been implicated in breast tumors of patients with shorter survival, suggesting that it may be a useful marker in identifying aggressive breast cancer.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For formalin-fixed paraffin-embedded immunohistochemical staining, a concentration range of 1 - 3 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.1 - 1 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

This antibody is effective in immunoblotting (WB) and immunohistochemistry (IHC).
