Total cell lysates (15 µg total protein) from the indicated cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-SKIL (SnoN) Antibody, clone O91H2, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

Description

Ski-like proto oncogene (SKIL) is a transcriptional repressor in the SMAD-TGF-β signaling axis that plays a critical role in TGF-β activation. It functions as a key regulator of proliferation and differentiation. Upon activation of TGFβ signaling, SMAD3 enters the nucleus and facilitates the ubiquitination and subsequent degradation of SKIL by the anaphase- promoting complex, resulting in derepression of SKIL-regulated genes and enhanced BMP-SMAD signaling. Additionally, SKIL is targeted for degradation by the E3 ligase Arkadia. The region corresponding to amino acids 1-96 of SKIL mediates its interaction with SMAD3, with residues 89-92 playing a critical role in stabilizing the interaction. The protein also contains a D-box motif between residues 162-174 that facilitates its interaction with other ubiquitin-conjugating enzyme complexes. Lysines 440, 446, and 449 are required for efficient ubiquitination upon TGF-β-activation.
