IHC staining of purified anti-Neurogranin antibody (clone A16007A) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with the primary antibody at 5 µg/mL overnight at 4°C. BioLegend's Ultra-Streptavidin (USA) HRP kit was used for detection followed by hematoxylin counterstaining, according to the protocol.

Lane 1: 20 µg of human brain tissue lysate; Lane 2: 20 µg of human Alzheimer's disease brain tissue lysate; Lanes 3 and 4: 5 µg of rat brain tissue lysate; Lane 5: 50 ng of human recombinant Neurogranin-GST protein. Blots were incubated with 1 µg/mL of the primary antibody overnight at 4°C followed by incubation with HRP labeled goat anti-mouse secondary antibody. Enhanced chemiluminescence was used as the detection system.

Description

Neurogranin (NRGN) is the main post-synaptic protein kinase substrate that binds to calmodulin (CaM) in the absence of calcium. It is suggested that NRGN is a direct target for thyroid hormone in the human brain, and that the control of expression of this gene could underlay many of the consequences of hypothyroidism on mental states during development as well as in adult subjects. Neurogranin in CSF has been implicated as a possible marker for synaptic degeneration in Alzheimer’s disease.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 - 5.0 µg per ml. For immunohistochemical staining on formalin-fixed paraffin-embedded tissue sections, the suggested use of this reagent is 1.0 - 5.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Cellular distribution: Golgi apparatus, mitochondria, nucleus, and cytoplasm in neuronal cell body and dendrites.

Function
Neurogranin is a neuron-specific, calmodulin-binding protein that participates in the protein kinase C signaling pathway. Neurogranin is a post-synaptic protein that modulates Ca2+ homeostasis in neurons by binding to and regulating the availability of calmodulin in the absence of Ca2+. Neurogranin function has been implicated in learning and memory.
