LEGENDplex™ is an assay platform using the Luminex xMAP® Technology for multiplex assays using unique sets of beads developed at Luminex. The assays built on the xMAP platform allow simultaneous quantification of multiple analytes in biological samples. BioLegend offers custom panels as well as custom target development for Luminex xMAP® assays.Click here for more information.
Most of our products (unless stated otherwise) have a guaranteed shelf life of one year from date of receipt by the end user, under proper storage and handling conditions as instructed on our Product Data Sheets.
For most products, we recommend storage undiluted at 4° C. Most Cell Biology products and lyophilized recombinant proteins are stored at -20° C. Frozen liquid recombinant proteins should be stored at -70° C (upon first thaw, aliquot and freeze at -70° C). Please refer to our Product Data Sheets for proper storage conditions.
Most products are shipped at ambient temperature. Recombinant proteins (including those in the ELISA MAXTM Sets) are shipped with dry ice in separate shipment.
YES! With an experienced biochemistry team, BioLegend is well positioned to offer a wide range of custom services to meet our customers' specific needs, as follows:
Antibody production: Cell culture, Purification Conjugation of BioLegend antibodies or customer's antibodies Fragmentation: F(ab')2, F(ab') Additional LEAFTM (Low Endotoxin, Azide-Free format) products for functional assays Alternate Antibody Concentration or Buffer
Antibodies offered per test have been pre-titrated for optimal performance in flow cytometry assays. Antibodies offered per plate have been pre-titrated in ELISA. Cell Biology antibodies offered per µl have been pre-titrated for Western Blot.
The antibody packed in an μg format (untitrated) is not necessarily always from the same batch number as a test format (pre-titrated) though the same procedures are performed in the quality control testing.
Please also note that not all test formats of the antibodies have lower concentrations than the μg format. It depends on a particular batch tested. If the optimal concentration were determined at 1 μg/test for a particular batch, then it would be 100 μg in the 100 tests.
The advantage to using a pre-titrated format is that it provides more convenience for the optimal performance in areas such as flow cytometry assays, especially for beginners who may save time and reagents on titration of the antibody.
100% Satisfaction Guarantee. If BioLegend's product does not perform as described on its product data sheet, we'll replace it or refund 100% of the original purchase price
For direct orders shipped outside the U.S., shipping costs typically start at USD $50 for antibody orders and $100 for dry ice shipment. This could vary country to country. Additionally, prepayment is required for direct international shipments. Also, taxes and duties will be paid by the recipient upon shipment delivery. In some countries, the customer needs to apply for an import permit. When ordering from a distributor, the payment terms are more flexible, the customer and technical support is more immediate, and the customer has greater convenience in not having to deal with customs agencies, duties/taxes, and currency exchange.
Check the Product Data Sheets on-line to review applications/characteristics for each clone. If you still have questions, please contact BioLegend Technical Services at:
BioLegend's LEAFTM (Low Endotoxin, Azide-Free) purified antibodies are specifically designed for functional analyses, providing the most accurate results with minimal negative effects. LEAFTM purified antibodies are tested for sterility, and endotoxin is <0.01 ng/µg of antibody.
For example, if you have a PE conjugated primary antibody with Rat IgG1, k isotype, use PE Rat IgG1, k isotype control Catalog # 400407 or 400408, as your isotype control. For your conveniece, most products have their isotype control listed under their "related products".
Instruction/Analysis Certificates are included with all ELISA MAXTM Sets. For other products, please contact BioLegend Technical Services for a Certificate of Analysis, and provide product's lot number.
For U.S. orders, if products are in-stock, they are shipped the same day, via Federal Express overnight, so you can expect to receive the products the next day. International direct orders are shipped on Mondays and Fridays, and typically arrive within 2 - 10 days, depending on how long they are in customs.
At the top right corner of our web page, click “Go” next to Product Search. This will take you to our product search function. Click the drop-down button to the right of “Keyword” and go to “Species Reactivity”. Enter one of the following species in the field, then click “Search”. All of the reagents that are reportedly cross-reactive with that species will be listed.
With a single 488 nm laser, three color staining can be performed with PE, FITC, and tandem dye PE/Cy5. For simultaneous use of APC, APC/Cy7, and PE-Cy7 dyes, multiple laser flow cytometers are needed. For more information, visit our Multicolor staining page.
In general our antibodies can be used for cell sorting, but we do not routinely test this in house. Please note the fluorochrome conjugates are not endotoxin tested and contain 0.09% azide. Typically this concentration is low, so the azide amount will not affect viability of the cells. As for activation of cells, typically the incubation time with the antibodies for staining is short (~20minutes) and this should not cause activation. Even if an antibody has functional activity (as viewed on the datasheet for LEAF formats) the concentration for functional activity will probably be higher than what it is for use in flow cytometry.
Both PE/Cy5 and PE/Cy5.5 are excited at 488 nm with an argon laser, but the emission of PE/Cy5 peaks at 670 nm and the emission of PE/Cy5.5 peaks at 690 nm. When used together with APC, PE/Cy5.5 requires less cross beam compensation than PE/Cy5.
The ELISA MAX™ Standard Sets contain pre-titrated Capture Antibody, Detection Antibody, Avidin-HRP Reagent, and Recombinant Standard.
The ELISA MAX™ Deluxe Sets contain all of the above, PLUS uncoated Plates, Coating Buffer, Assay Diluent, and TMB Substrate Reagent.
We also have the LEGEND MAX™ kits which are ELISA kits with Pre-coated Plates, pre-titrated Capture Antibody, Detection Antibody, Avidin-HRP Reagent, Recombinant Standard, Coating Buffer, Assay Diluent, and TMB Substrate Reagent. These ready to use reagents are provided with full analytical validation.
No, the plates are uncoated, but Coating Buffer and Assay Diluent (for blocking and dilutions) are included in the ELISA MAXTM Deluxe Sets. Instructions on coating the plates are in the Sets' product manuals and website under "Support".
This is not recommended. The ELISA MAXTM reagents are optimized for a particular set lot, and they are not recommended for applications other than ELISA.
We recommend adding 3x105 to 1x106 cells per well, depending on the experiment. If cells are added prior to the antigen or mitogen, the spots may be distributed unevenly, to the edges of the wells.
Avoid air bubbles when coating the plates, as this may cause blank spots on the wells. Coated plates should be stored overnight at 4° C in a humidified box. To avoid contamination, maintain sterile techniques during the coating and incubation periods.
Plates should be washed by adding wash solution (without touching the membrane on the well bottoms), then flicking it out. To avoid damage to the membrane, do not use an automated plate washer.
Transfer efficiency may have been poor. Check protein transfer by staining the gel and/or membrane.
Incorrect storage of antibodies or ECL western blotting detection reagents.
Insufficient protein may have been loaded on the gel. Depending on the location of the target protein, membrane or nuclear preparations may be required (instead of whole cell lysates).
