Product Details

Source: Rat IL-6, amino acids Phe25-Thr211 (Accession # NM_012589), was expressed in E. coli.
Formulation: 0.22 µm filtered protein solution is in 38% Acetonitrile, 0.1% TFA.
Endotoxin Level: Endotoxin level is <0.1 EU/µg (<0.01ng/µg) protein as determined by the LAL method.
Preparation: 10-100µg sizes are bottled at 200µg/mL. 500µg sizes and larger are bottled at the concentration indicated on the vial.
Storage & Handling: Unopened vial can be stored at 4°C for three months. For maximum results, quick spin vial prior to opening. Stock solutions should be prepared at no less than 10µg/mL in buffer containing carrier protein such as 1% BSA or HSA or 10% FBS. Do not freeze. Freezing has been shown to reduce bioactivity.
Activity: ED50 is < 0.01 ng/ml, corresponding to a specific activity of > 1 x 108 units/mg, as determined by the dose dependent stimulation of 7TD1 cell proliferation.

Description:

The cDNA sequence coding for the mature portion of rat IL-6 is 92% identical with the corresponding murine sequence and 68% identical with the corresponding human sequence. The two rodent IL6 species are highly conserved. IL-6 is a multifunctional cytokine with both differentiation and growth-promoting effects for a variety of target cell types. It is a B-cell stimulatory factor inducing terminal differentiation and high level antibody production in B lymphocytes (IL6/BSF-2) and it acts as a hybridoma/plasmacytoma growth factor. IL-6 stimulates the growth of hemopoietic stem cells, and induces the synthesis of acute phase plasma proteins (2). In addition, IL-6 and tumor necrosis factor alpha (TNF-alpha), involved into osteoclastogenesis and responsible for bone resorption process, participate in the pathogenesis of the osteoporosis (3). IL-6 plays a pivotal role in liver regeneration (4). It is systemically elevated in obesity and is a predictive factor to develop type 2 diabetes, and regulates pancreatic alpha cell mass expansion (5).

Function: Upregulated by IL-1, PDGF, IFN-β, TNF-α, NGF, IL-17; downregulated by glucocorticoids IL-4, TGF-β
