Source: Mouse IL-25, amino acids Val17-Ala169 (Accession # NM_080729) was expressed in E. coli.
Endotoxin Level: Endotoxin level is <0.1 EU/µg (<0.01 ng/µg) protein as determined by the LAL method.
Preparation: 10-100 µg sizes are bottled at 200 µg/mL. 500 µg sizes and larger are bottled at the concentration indicated on the vial.
Concentration: 0.22 μm filtered protein solution is in 20 mM NaHPO4 pH 6.0, 0.5M NaCl
Storage & Handling: Unopened vial can be stored at 4°C for three months or -20°C to -70°C for six months. For maximum results, quick spin vial prior to opening. Stock solutions should be prepared at no less than 10 µg/mL in buffer containing carrier protein such as 1% BSA or HSA or 10% FBS. Stock solution can be stored up to 3 months from -20°C to -70°C under sterile conditions. Avoid repeated freeze/thaw cycles.
Activity: Mouse IL-25 binds to recombinant mouse IL17BR/Fc Chimera in a functional ELISA.

Description:

IL-25E (also known as IL-17E) is a member of the IL-17 family, which includes IL-17A, IL-17B, IL-17C, IL-17F and IL-17A/F. Primary sequence homology differs between family members, with hIL-17 and hIL-17F having the highest homology (44%), while hIL-17 and hIL-25 have the lowest (15%). IL-25 induces elevatedgene expression of IL-4, IL-5  and IL-13 in multiple tissues and results in T helper 2 (TH2)-type immune responses (increased serumIgE levels) and pathologic changes in the lungsand digestive tract with eosinophilic infiltrates, increased mucusproduction, epithelial cell hyperplasia and overall amplified allergicinflammation. IL-25 shares the receptor IL-17RB with IL-17B, although it binds with much higher affinity than IL-17B.

Structure: Heterodimer
Distribution: IL-25 is produced by mast cells, epithelial cells, alveolar macrophages, microglia and Th2 cells.
Function: IL-25 directly promotes Th2 cell differentiation. IL-25 induces the expression of IL-4, IL-5, IL-13, and eotaxin and produces lung eosinophilia. In addition, IL-25 treatment of differentiated Th9 cells enhanced IL-9 production.
Ligand Receptor: IL-25 binds to IL-17RB
Interaction: CD4+ NKT cells, macrophages, endothelial cells, eosinophils, Th2, and Th9 cells.
