Extracts of untreated HepG2 cells (lane 1) and extracts of HepG2 cells treated with 300 nM of thapsigargin for 16 hours (lane 2) were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified polyclonal anti-Xbp-1 antibody (clone Poly6195). The proteins were visualized using a donkey anti-rabbit-IgG secondary conjugated to HRP and chemiluminescence detection.
Xbp-1 (X-box binding protein 1) is a ubiquitously expressed 29 kD member of the CREB/ATF transcription factor family, and a mammalian homologue of the yeast transcription factor Hac1. Xbp-1 regulates MHC class II genes by binding to the X-box promoter motif and can also bind to the enhancer in T cell leukemia virus type I promoter to increase viral protein expression by binding the viral transactivator. This transcription factor is essential for hepatocyte differentiation and can be upregulated by ER stress, IL-6, and IL-4. The PAX5 transcription factor downregulates Xbp-1; downregulation has been shown to correlate with tumor progression in prostate cancer. The Poly6195 antibody recognizes the C-terminal region of human and rat Xbp-1 and has been shown to be useful for Western blotting.
Other Names:
X-box binding protein 1, XBP1
Structure:
Member of the CREB/ATF transcription factor family, mammalian homologue of the yeast transcription factor Hac1. Predicted molecular weight approximately 30 kD and 54 kD (splicing variant)
Distribution:
Ubiquitously expressed
Function:
Transcription factor regulating MHC class II genes by binding to X-box promoter motif. Can also bind to enhancer in T cell leukemia virus type I promoter and enhance viral protein expression by acting as DNA binding partner of a viral transactivator.
Regulation:
Upregulated by ER stress, IL-6, and IL-4. Downregulation correlates with tumor progression in prostate cancer. Downregulated by PAX5 transcription factor
