Each lot of this antibody is quality control tested by Western blotting. Western blotting, suggested working dilution(s): Use 10 µl per 5 ml antibody dilution buffer for each mini-gel. For IHC, use a 1:50 dilution of antibody for staining. Antigen retrieval for IHC of formalin-fixed paraffin-embedded tissue using 0.01 M sodium citrate buffer is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Formalin-fixed paraffin-embedded human tonsil tissue was stained with Poly6248 and developed with an alkaline phosphatase chromogen substate (red color). Tissue was counterstained with H&E (blue/pink). Magnification, 40X.
Hela cell extract was resolved by electrophoresis, transferred to nitrocellulose, and probed with rabbit anti-Sp3 polyclonal antibody. Proteins were visualized using a donkey anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence system.
The sequence-specific transcription factor Sp3 binds GC and GT box sequences in the promoters of a variety of genes. The Sp3 transcription factor contains three zinc finger domains that determine sequence specificity of DNA binding. This transcription factor is thought to regulate nucleoside, nucleotide and nucleic acid metabolism and many other "housekeeping" genes. Sp3 can be modified by sumoylation by Ubc9 and has been shown to interact with other transcription factors and histonbe deacetylases. Sp3 has been reported to act as a transcriptional enhancer and repressor of Sp1 activity in a cell- and context-dependent manner. The Poly6248 antibody recognizes the C-terminal region of human, mouse, and rat Sp3 and has been shown to be useful for Western blotting.
