Recommended Usage: Each lot of this antibody is quality control tested by Western blotting. Western blotting, suggested working dilution(s): Use 10 μl per 5 ml antibody dilution buffer for each mini-gel. It is recommended that the reagent be titrated for optimal performance for each application.
A431 cell extract was resolved

A431 cell extract was resolved by electrophoresis, transferred to nitrocellulose and probed with rabbit anti-PTEN antibody. Proteins were visualized using a donkey anti-rabbit secondary conjugated to HRP and a chemiluminescence detection system. This polyclonal antibody recognizes two specific bands at approximately 47 kD and 60 kD.


Description: PTEN (also known as phosphatase and tensin homolog deleted on chromosome ten and MMAC1) is a protein-tyrosine phosphatase non-receptor containing a tensin domain and PDZ binding domain. This 47 kD protein is found in the cytoplasm and functions as a potential tumor suppressor. PTEN is a negative regulator of the PI3K/AKT signaling pathway regulating phosphoinositide(3,4,5)P3 levels, and an inhibitor of cell migration. PTEN is mutated in numerous cancers. PTEN activity is inhibited by phosphorylation and is transcriptionally upregulated by PPARγ agonists. PTEN has been shown to interact with phosphatidylinositol 3,4-bisphosphate, PtdIns(3,4,5)P(3), MAGI-1, MAGI-2, MAGI -3, hDLG, and MAST205. The Poly6144 antibody recognizes the N-terminal region of human PTEN and has been shown to be useful for Western blotting and immunofluorescence staining.
