This antibody is provided in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 50% glycerol.
Preparation:
The antibody was purified by antigen-affinity chromatography.
Storage & Handling:
Upon receipt, store frozen at -20° C.
Application:
WB
Recommended Usage:
Each lot of this antibody is quality control tested by Western blotting. Western blotting, suggested working dilution(s): Use 10 μl per 5 ml antibody dilution buffer for each mini-gel. It is recommended that the reagent be titrated for optimal performance for each application.
Hela cell extract was resolved by electrophoresis, transferred to nitrocellulose and probed with rabbit anti-p130 Cas antibody. Proteins were visualized using a donkey anti-rabbit secondary conjugated to HRP and a chemiluminescence detection system.
Description:
p130 Cas (Crk-associated substrate) is a 120-130 kD docking protein and a central coordinator for tyrosine-kinase-based signaling related to cell adhesion. p130 Cas has been implicated in induction of cell migration, growth factor stimulation, cytokine receptor engagement, cardiovascular development, and actin filament assembly. p130 Cas is localized to focal adhesions and stress fibers. When unphosporylated, localizes in the cytoplasm; phosphorylation by FAK induces movement to membrane. p130 Cas is dephosphorylated by PTP-PEST, dephosphorylation blocks cell migration. p130 Cas has been reported to interact with v-Crk, v-Src, nephrocystin, PTK2B, CrkII and to form complexes with Fak1, CrkL, and Lyn kinase and to heterodimerize with CasL. The Poly6139 antibody recognizes the C-terminal region of human p130 Cas and has been shown to be useful for Western blotting.
Focal adhesions, stress fibers; unphosphorylated localizes to cytoplasm, moves to membrane upon tyrosine phosphorylation
