IL-17 or IL-17A originally identified as cytotoxic T lymphocyte-associated antigen-8 (CTLA-8) in mouse, is a founding member of the IL-17 family of cytokines. IL-17 is mainly produced by a small subset of T helper cells called TH17 cells. IL-17 is a 155 amino acid protein that is a disulfide linked, homodimeric, secreted glycoprotein with a molecular mass of 35 kDa. The amino acid sequence of human IL-17 is 63% and 58% identical to mouse and rat IL-17, respectively, and 72% identical to the ORF13 of Herpesvirus saimiri. IL-17 exhibits multiple biological activities including the induction of various proinflammatory cytokines and chemokines such as IL-6, IL-8, G-CSF, GM-CSF, GRO- α, and MCP-1 on a variety of cell types. IL-17 is also involved in the activation of NF-kB and costimulation of T cell proliferation. As a result of these roles, IL-17 has been linked to many immune/autoimmune related diseases including rheumatoid arthritis, asthma, lupus and multiple sclerosis.
